The Yes-associated protein (YAP) oncoprotein is linked to metastasis and chemotherapy resistance in lung cancer cells. This study explored the effects of inhibiting ๐๐๐ ๐ฎ๐ฌ๐ข๐ง๐ ๐๐๐/๐๐๐ ๐๐จ๐ฆ๐๐ข๐ง (๐๐๐๐) ๐ญ๐ซ๐๐ง๐ฌ๐๐ซ๐ข๐ฉ๐ญ๐ข๐จ๐ง ๐๐๐๐ญ๐จ๐ซ ๐ข๐ง๐ญ๐๐ซ๐๐๐ญ๐ข๐จ๐ง inhibitors in chemo-resistant lung cancer cells.
The authors examined YAP activation, cell migration, and TEAD transcriptional activity in ๐๐๐ง๐๐๐ซ ๐๐๐ฅ๐ฅ ๐ฅ๐ข๐ง๐๐ฌ with elevated YAP levels. They analyzed parental (A549) and paclitaxel resistant (A549R) cell lines. A ๐๐ ๐ฆ๐ข๐๐ซ๐จ๐๐ฅ๐ฎ๐ข๐๐ข๐ ๐๐ฎ๐ฅ๐ญ๐ฎ๐ซ๐ ๐๐๐ฏ๐ข๐๐ was used to study a YAP/TEAD inhibitor (IV#6)/paclitaxel combination's effect on cells from fresh lung cancer samples.
Results showed ๐ก๐ข๐ ๐ก๐๐ซ ๐๐๐ ๐๐๐ญ๐ข๐ฏ๐ข๐ญ๐ฒ in resistant cells, with YAP/TEAD inhibitors reducing YAP activity and cell motility. ๐๐จ๐ฆ๐๐ข๐ง๐ข๐ง๐ ๐๐๐ ๐ข๐ง๐ก๐ข๐๐ข๐ญ๐ข๐จ๐ง ๐ฐ๐ข๐ญ๐ก ๐ญ๐ซ๐๐ฆ๐๐ญ๐ข๐ง๐ข๐ ๐จ๐ซ ๐ฉ๐๐๐ฅ๐ข๐ญ๐๐ฑ๐๐ฅ sensitized resistant cells and ๐ค๐ข๐ฅ๐ฅ๐๐ ๐๐๐ง๐๐๐ซ ๐๐๐ฅ๐ฅ๐ฌ in a 3D system. This study highlights the potential of targeting YAP/TEAD to overcome chemotherapy resistance.
